A major challenge in the use of HepG2 cell culture models for drug toxicity screening is their lack maturity 2D culture. 3D Matrigel promotes formation spheroids that express liver-relevant markers, yet they still various primary hepatocyte functions. Therefore, alternative matrices where chemical composition and materials properties are controlled to steer maturation remain desired. Herein, a modular approach taken based on fully synthetic minimalistic supramolecular matrix squaramide synthons outfitted with cell-adhesive peptide, RGD spheroid Co-assemblies RGD-functionalized squaramide-based native monomers resulted soft self-recovering hydrogels tunable concentration. self-assembled grown (≈150 µm) within high viability differentiation over 21 days Importantly, significantly higher mRNA protein expression levels phase I II metabolic enzymes, transporters, liver markers found comparison Matrigel.
